Líneas de Investigación

En colaboración con estudiantes (pre y post-grado), investigadores Chilenos e internacionales, estudiamos la regulación y papel funcional de canales de membrana formados por conexinas (Cxs) o panexinas (Panxs), que son proteínas expresadas por la mayoría de las células de los vertebrados. Dichas proteínas constituyen canales denominados canales de uniones en hendidura (CUH, "gap junction channels") y hemicanales (HCs) o conexones. Los CUH comunican el citoplasma de células en contacto y los HCs comunican el medio intra con el extra celular. También estudiamos la interacción entre canales formados por Cxs o Panxs y los receptores purinérgicos (P2) activados por ATP extracelular. Todos estos canales juegan un papel crucial en la coordinación de numerosas respuestas celulares en diversos tejidos. Nuestros estudios se centran principalmente en el funcionamiento normal y aberrante de células de los sistemas nervioso e inmune. Trabajamos, además, en diversas otros tipos celulares incluyendo los músculos esqueléticos. Utilizamos cultivos celulares primarios, líneas celulares y modelos animales de enfermedades inflamatorias o que evolucionan con una respuesta inflamatoria. Estudiamos el efecto de neurotransmisores, hormonas y condiciones pro-inflamatorias sobre el grado de comunicacion intercelular y sobre el grado de comunicación intercelular y la extensión de muerte celular mediada por los canales de interés. Utilizamos técnicas bioquímicas, biofísicas, celulares y moleculares para evaluar la expresión, modificaciones post-traduccionales, y tráfico intracelular de las Cxs y Panxs. Utilizando técnicas electrofisiológicas y de imágenes evaluamos cambios de conductancia y permeabilidad de los canales formados por dichas proteínas. En colaboración con industrias (I D) estamos desarrollando compuestos que afectan estos canales con el fin de controlar los efectos nocivos inducidos por condiciones pro-inflamatorias. Ya que la regeneración es la última etapa de la respuesta inflamatoria y esta comparte numerosas características con la ontogenia de los tejidos, también estamos desarrollando investigación sobre la regulación y función de los canales de interés en el crecimiento y la diferenciación celular. La finalidad de lo último es adquirir más conocimiento sobre el papel de los CUH y HCs en la tumorogénesis.
