Líneas de Investigación

1.-  Mecanismos moleculares de la respuesta al estrés celular en neuronas.

Las neuronas tienen una respuesta disminuida al estrés celular. La razón de este fenómeno es desconocida. Nosotros mostramos que el co-represor transcripcional CoREST regula la respuesta a estrés celular en líneas celulares no neuronales. CoREST forma un complejo represor transcripcional con la desmetilasa de histonas, LSD1 y las desacetilasas de histonas (HDACs 1/2). Estamos estudiando el rol del complejo represor CoREST/LSD1/HDACs y del heat shock factor 1 en la respuesta a estrés celular en cultivo primario de neuronas corticales y en el modelo de diferenciación de células PC12 por NGF.

2.- Rol del receptor nuclear Nurr1 en la mantención del fenotipo neuronal dopaminérgico.

Las neuronas dopaminérgicas mesencefálicas juegan un rol fundamental en las conductas motivadas y la adicción a drogas, y en el control de los movimientos voluntarios, entre otras funciones. El receptor nuclear huérfano Nurr1 es esencial en el origen y mantención de estas neuronas dopaminérgicas. Nosotros hemos determinado que la sumoilación y la SUMO E3 ligasa PIASg regulan la actividad transcripcional de Nurr1. Utilizando un virus adeno-asociado codificante de una ribozima contra Nurr1, mostramos in vivo que Nurr1 regula la expresión de RET, el receptor de GDNF. Actualmente, estamos llevando a cabo experimentos para entender a nivel molecular cómo Nurr1 regula la expresión de RET y de qué manera influye la sumoilación en el control que ejerce Nurr1 sobre sus genes blanco. 

3.- Modificaciones neuroquímicas y moleculares en un modelo del desorden obsesivo-compulsivo (OCD).

En ratas, la administración repetida de agonistas dopaminérgicos D2 induce una conducta similar al desorden obsesivo-compulsivo. En este modelo experimental de OCD estudiamos cómo se modifican la neurotransmisión dopaminérgica y glutamatérgica en el núcleo accumbens y cuáles son los cambios que ocurren en la expresión de genes en este núcleo. En particular nos enfocamos en los factores de transcripción Nurr1 y Nur77 que se expresan en las neuronas dopaminérgicas y dopaminoceptivas, respectivamente.
