Líneas de Investigación

La enfermedad de Alzheimer (EA) es una demencia progresiva que no tiene tratamiento efectivo. Considerando los cambios neuropatológicos, se han propuesto varias hipótesis que podrían explicar la enfermedad: Nuestro interés es la "hipótesis de la cascada del amiloide",  en la cual el proceso neurodegenerativo presente en cerebros de pacientes con EA se explicaría por el procesamiento anormal de la proteína precursora del ß-amiloide (APP), cuya consecuencia es la producción, agregación, deposición y toxicidad del péptido ß-amiloide (Aß). Esta hipótesis ha recibido apoyo a partir de estudios genéticos de la forma familiar de la EA de aparición temprana, en la cuál se ha mostrado que mutaciones en el APP o en los genes de las presenilinas (PS1 y PS2), determinan un incremento en la producción de Aß, por mecanismos aún no definidos. A pesar que estos estudios, han probado ser significativos en la comprensión del inicio de la forma familiar (1%), algunas preguntas permanecen sin respuesta: por ejemplo, (1) cuáles son los mecanismos moleculares involucrados en el inicio de la forma esporádica de la enfermedad (99%), y (2) cual es la razón de la vulnerabilidad selectiva de las áreas del cerebro afectadas, tales como el hipocampo y la corteza entorrinal o el sistema colinérgico del cerebro anterior.

Un aspecto importante en los cerebros de pacientes con EA, es la disfunción sináptica en la cual se produce una alteración primaria en la region post-sináptica y luego en la pre-sináptica, lo que se explicaría por la toxicidad de agregados del péptido Aß, llamados oligómeros de Aß los cualesl se acumulan en la región post-sináptica de las espinas dendríticas, donde ejercen efectos que llevan a las alteraciones cognitivas de la EA. Actualmente estamos estudiando el papel de la vía Wnt de señalización en la estructura y función sináptica, junto a su papel en los mecanismos moleculares responsables de la deposición del amiloide intracelular, como así también en la plasticidad sináptica.

En una línea biotecnológica e innovativa, hemos identificando compuestos que esperamos permitan combatir  la EA, en particular algunos productos naturales y/o activadores de la vía Wnt a nivel cerebral, utilizando ratones doble transgénicos (APPswe PS-1) modelos del Alzheimer.
